The major goal of the Immunology Core is to provide flow cytometry technical support and expertise to
facilitate studies in HIV/AIDS research. In addition to general technology, the Core will provide unique
services and expertise for studies that analyze quantitative changes in T cell subsets. In the support of studies
being done-by CFAR investigators_ specifi_ Aims of the eore inelude: (t) to provide machinery and technical
support for studies requiring the analysis of cell markers using flow cytometry; (2) to provide tools and services
and expertise for studies that quantitate T cell subsets expressing different T cell receptor (TCR) V regions and
that quantitate antigen-specific T cell subsets by flow cytometry; and (3) to provide machinery and technical
support for studies requiring the sorting of potentially infected cells by flow cytometry. Aim 2 will include
analyses of TCR V region expression, staining with major histocompatibility complex (MHC)/peptide
multimers, and T cell intracellular cytokine expression after stimulation with antigen. To support Aims 2, the
Core will maintain a panel of monoclonal antibodies directed to different TCR V regions and provide quality
control for the use of these reagents. In addition, the Core will produce and monitor MHC class II/peptide
multimers for CFAR investigators to use for analysis of antigen-specific T cells relevant to HIV infected
individuals. Support of the Immunology Core will allow the use of these reagents and performance of analyses
based on flow cytometry in a consistent and high quality manner.
