RIXS STUDIES OF PLATINUM COMPLEXES FOR CANCER THERAPY

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Cisplatin (cis-[PtCl2(NH3)2])) is one of the most widely used anticancer drugs and has been shown to be an effective treatment for ovarian, testicular and cervical cancers. However, despite the fact that it is highly effective in treating several types of cancer; cisplatin is also toxic and there are many types of cancer that it does not effectively target. In an effort to improve treatment, much research has focused on synthesizing different type of Pt complexes with reduced toxicity, increased activity, and the ability to be administered orally. Many of these small chemical compounds can be crystallized and characterized by diffraction studies. However, in order to understand the mechanism of these complexes, an understanding of the solution structure is vital. Recently we have used Pt L3-edge XAS to characterize solid and solution structures of Pt anticancer agents, including Pt-Streptidine. Future studies will include examination of the Pt coordination environment within cancerous cells.
