STRUCTURAL STUDIES ON MAMMALIAN AND BACTERIAL BINDING PROTEINS AND ENZYMES

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The general area of research in our laboratory involves the determination of the 3-D structures of proteins and enzymes of biomedical and industrial importance. This renewal for beam time on BL7-1 and BL9-2 at SSRL focuses on the collection of high resolution diffraction data from mammalian and bacterial binding and transport proteins, along with a number of enzymes involved in folate metabolism, bacterial cell wall biosynthesis, antibiotic resistance and disulfide bond isomerization. The desire to collect data at SSRL is based upon our need for high quality, high resolution diffraction data from a number of different crystals, some of which are either too small or sensitive for conventional x-ray sources, or have long cell edges. In addition, we are now producing the majority of our proteins as seleno-methionine derivatives, and we envisage an increased requirement for the collection of accurate MAD data.
