This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. Work in the budding yeast Saccharomyces cerevisiae has uncovered a synthetic lethal genetic interaction network for genome stability genes, a few members of which have interactions with many other members of the network. One of these central genes is CTF4; it and many of its genetic interacting partners are conserved across evolution. Many of the human homologs of CTF4's genetic interactors have been previously shown to be mutated in human cancers. Probing the physical interactions of the Ctf4 protein would serve to fill out its interaction network, and may also reveal additional conserved targets for anticancer therapy. Interactors identified in a yeast two-hybrid screen will subsequently be confirmed by reciprocal two-hybrid interaction and co-immunoprecipitation/mass spectrometry.
