This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Ty3 produces virus-like particles similar to the core particles of retroviruses. In the case of Ty3 however, about 60 Ala scanning mutations in the structural protein open reading frame are available with which to study interactions of the structural protein with itself and with host proteins. We have constructed Gag3 insertions in two hybrid bait and activation vectors which target Gal4-mediated control. The structural protein (Gag3) has been shown to interact with itself but to be affected in certain mutants. Two of the mutations are candidates for affecting interactions between Gag3 and host proteins. Preliminary data suggests that these proteins might be involved in vesicular trafficking and P-body localization. We propose to screen a yeast genomic library in order to test for host proteins that interact with Ty3 Gag3 and are involved in particle assembly.
