This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
LONG TERM GOAL: Obtain funding for a randomized controlled trial, to test a cancer preventive agent. This study will obtain pilot data that will help achieve this goal.
SPECIFIC AIMS: Smaller low density lipoprotein (LDL) molecules are less resistant to oxidative modification, which appears to increase general markers of oxidative stress in the body. Oxidative stress is hypothesized to play a key role in prostate cancer. Correlating LDL particle size with prostate cancer would be important, since medications exist that can increase LDL size. This case control study will compare LDL particle size, markers of oxidative stress, and serum lipoprotein levels in prostate cancer patients with matched controls.
HYPOTHESIS: Prostate cancer patients will have significantly smaller LDL particles than age and risk factor matched controls.
STUDY POPULATION: This study will target adult men between 40-80 years of age.
EXCLUSION CRITERIA: Metastatic Prostate Cancer, Cancer other than prostate cancer, hemochromatosis.
DESIGN: This study will consist of a case control design with control patients that will be matched on age and current smoking status.
CASES: Prostate cancer cases will be generated from Dr. Klein's urology practice at the CCF.
CONTROLS: Controls will be recruited through the Department of Preventive Oncology at the CCF main campus. Controls will be matched on age and current smoking status. Control patients will consist of individuals without a history of cancer. Other controls variables will include: race, socioeconomic status, hypertension, diabetes, smoking, and use of a lipid lowering medication.
SAMPLE SIZE: To achieve a power of 80% at detecting an effect size of 0.5 standard deviation difference in LDL particle size between the groups with a one tailed test and and alpha of 0.05, will require a sample size of 120 participants. Patients will be recruited in 2:1 ratio of controls:cases.
ANALYSIS: LDL particle size will be compared between the cases the controls using Multiple Analysis of Variance with covariates.
