RADIATION DAMAGE AS A FUNCTION OF WAVELENGTH

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. In order to fine-tune data collection parameters in two-wavelength MAD experiments, we also seeked to determine how radiation damage affects the data at different energies over the absorption edge. To do this we collected data from different lysozyme crystals derivatized with Galeodinium. Successive data sets were collected at four different points: on the low energy side of absorption edge, inflection point, absorption peak and on the high energy side of the edge. We examined cysteine Sulphur reduction and the Wilson temperature factor as the indicators of radiation damage. It was difficult to conclude from the data whether a different amount of specific damage was observed at different energies; The temperature factor increases at lower energies, which suggests that the dose on the crystal is not the only factor affecting the amount of damage suffered by the crystals.
