This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Developing digital imaging systems for intermediate voltage electron microscope (IVEM) is the objective of this core project. Previously, we discussed the installation of a new retractable 1K x 1K fast camera for the JEM-3200F. This Fast-scan camera was installed on the JEM-3200FX and is providing excellent images. We report this year on a new installation of the 1k x 1k, F114-R fast scan CCD from Tietz Video and Image Processing Systems (TVIPS) on the resource JEM-4000EX. This camera features a 12 frames-per-second readout at full resolution (25 frames-per-second, binned 2 x 2), 12 bit analog-to-digital converter, delivering greater that 12% contrast at Nyquist. One of the most useful features of this system is the ability to provide nearly real time fast Fourier transforms, providing unmatched feedback for correcting astigmatism. With the improved performance of this wide-angle implementation, the IVEM is well equipped to deliver large format, high quality video feedback in support of telemicroscopy and automated tomography (e.g. FasTomo, etc.). In the past year, we have integrated the fast-scan camera into NCMIRs Generalized Telemicroscopy suite of control software. With much of the software integration and development completed around the JEM-3200 instrument, we anticipate the integration of the new F114-R on the JEM-4000EX system to be quick.
