This subproject is one of many research subprojects utilizing the

Mitotic spindle function critically depends on the kinetochore, a proteinacous structure that serves to connect spindle microtubules to centromeric DNA. While numerous kinetochore proteins have been identified in diverse organisms, how these proteins are organized to connect chromosomes to spindle microtubules, and whether any structural and organizational themes are common to kinetochores of distantly related organisms, are key unanswered questions. Here, we use affinity chromatography and mass spectrometry to generate a map of kinetochore protein interactions. Phosphorylation sites of kinetochore proteins are mapped and the functional consequences of mutating these sites are analyzed in vivo. This will allow a deeper understanding of the architecture and regulation of the budding yeast kinetochore.
