英文摘要
This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This project aimed to use the unique capabilities of our recently developed ESI-qQq-FTMS to fully characterize the post-translational modifications of H-RAS that occur during oxidative stress conditions. The project used a combination of techniques including top-down and bottom-up proteomics methods, selective accumulation, Q2-CAD, and ECD. The samples involved are commercially purchased H-RAS, both native state and oxidatively modified with peroxynitrite. This was done in-vitro initially, but the goal is to extend this to in-vivo models once the in-vitro modifications are characterized. A paper was published in Analytical Chemistry on this work.
