项目摘要

Abstract The Imaging Core (Core C) will provide services related to sample processing, image acquisition and image analysis for all PPG investigators. Due to the unconventional size and intricate polymeric organization of mucins, studying complex biological systems like mucus require innovative, workable, and reliable experimental approaches for proper investigations. Core C offers expertise in mucus-rich sample handling/processing/staining and technical proficiency for image acquisition using a broad range of microscopy techniques. Imaging techniques available through Core C include RNA-in situ hybridization, immunohistochemistry, mucin agarose Western blotting, scanning and transmission electron microscopy, and atomic force microscopy. The combination of all of these imaging techniques will provide alternative ways to visualization key components of the mucociliary apparatus and gain insights into the molecular interactions that govern the viscoelastic properties of mucus. In addition to providing high-quality micrographs, Core C assists investigators with state-of-the-art image analysis using various software, (e.g., ImageJ, VisioPharm, Odyssey) to ensure accurate quantitative analyses of digital images. Indubitably, the proper functioning of Core C relies on partnerships established for years between the Ehre lab and existing microscopy facilities (MSL and HIC) on UNC campus. The use of these recharge centers and their existing equipment has been carefully allocated in the budget section. Core C is designed to provide protocols, reagents, as well as expertise, guidance and technical assistance for sample processing, image acquisition and digital image analysis for projects 1, 2, 3, and 4.
