Abstract Supplementary funds are requested to purchase a glow discharge apparatus to be used nearly daily for the projects funded by this R35. Glow discharge of EM grids is needed for both negative stain and cryo-EM applications to clean the grids and make them more hydrophilic. This is a very important step in sample preparation, and is crucial to getting good ice for cryo-EM. The existing apparatus was purchased in my lab ~ 13 years ago, is failing to function properly, and cannot be repaired in any cost-effective way.
