SERUM ORGANOCHLORINE COMPOUNDS AND BREAST CANCER ON LI

We are carrying out an NCI-funded case control study of breast cancer in relation to environmental and lifestyle factors (CA-63021) in two large Long Island hospitals as part of the Federally mandated Long Island Breast Cancer Study Project. A major aim of this study is the evaluation of breast cancer risk in relation to prior exposure to certain organochlorine compounds (OCC), including pesticides such as DDT and its metabolites, as well as polychlorinated biphenyls (PCBs). Exposures are evaluated via a structured questionnaire and through chemical analysis of samples of adipose tissue obtained surgically from patients and controls. At the time the grant was submitted, nearly most epidemiological literature on OCC in relation to breast cancer was based on adipose tissue. Recently, several important studies have been published which make an extension to serum both desirable and urgent. With IRB approval and informed consent from all patients, we have obtained serum samples along with adipose tissue, and have accumulated an archived serum bank of approximately 600 frozen sera out of an eventual study goal of 900 samples (300 cases, 300 benign breast controls, and 300 surgical controls). All serum samples were collected prior to treatment. This proposal has the following aims: 1. To assay the archived serum samples for the same OCC analytes (pesticides and PCB?S) that are now assayed in adipose tissue. 2. To study the relationship between adipose and serum levels of OCC within individual subjects in order to improve the interpretation of studies which use only one (usually serum ) of these biological sources. 3. To study the relationship between adipose and serum levels of OCC and other breast cancer risk factors, such as age, body mass index, education, and residence. 4. (a) To estimate the risk of breast cancer in relation to serum levels of OCC adjusted for other risk factors, (b) to determine whether such estimates differ materially from those based on adipose levels, and if so (c) to determine reasons for such differences.
