In this proposal I request a digitally controlled microinjection system for labeling cells in embryos of sponges and following their fate in the adult body. The requested equipment will allow students to study the specific activities of individual cells using fluorescent markers and follow their position in the embryos and juvenile animals with fluorescent imaging techniques and 3D reconstruction software. A highly controlled method of locating the cells is needed as well as a precise method of applying the dye. Attempts to do this without this equipment have failed due to the imprecision and rough movement of manual manipulators. The new instrument will significantly improve ease of use by my graduate students and associates thereby increasing both user-time efficiency and training.
