英文摘要
In this proposal I request funds for an oxygen microsensor system for simultaneous recording of oxygen levels in ambient water and water filtered by sponges. The requested equipment will allow students to study the metabolism of sponges under different ambient conditions (low and high flow) and in response to pharmacological and gene knockdown treatments. An accurate, stable and robust method of quantifying oxygen use by the sponge is needed, as is the ability to record from control and experimental locations simultaneously. Work with borrowed instruments has shown how valuable this data is for calculating energetics of sponge function, but also highlighted difficulties with earlier technologies in terms of drift and fragility of probe tips. The proposed instrument uses new very stable technology that has proven very effective. This instrument is urgently needed for specific experiments of 3 graduate students and their results will be used by two other members of the research group for modeling. The ability to use this microsensor will enhance productivity for all my group members.
