The principal applicant for this RTI application is Dr. David Wright with Drs. David Dyck, Jeremy Simpson and Amanda Wright serving as co-applicants. Dr. D.Wright's laboratory examines the mediators of exercise and diet on the regulation of adipose tissue and liver function and metabolism. The main objective of Dr. Wright's basic research program, which is funded through an NSERC Discovery Grant, is to elucidate the role and underlying mechanisms of the hormone glucagon in controlling the secretion of liver derived hormones called hepatokines, and, in turn, examining how these alterations impact whole body carbohydrate and lipid metabolism. The analysis of hepatokine gene expression is a central outcome measure of these experiments. Dr. D.Wright's research dovetails with the on-going NSERC-funded investigations in Dr. Dyck's research laboratory which focuses on the basic mechanisms controlling the uptake, oxidation and storage of carbohydrate and lipid in skeletal muscle and how these intricate processes are regulated by adipose tissue derived signaling factors called adipokines. The mechanistic, metabolic research programs of Drs Dyck and D.Wright complement Dr. Jeremy Simpson's NSERC funded program that examines the post-translational modifications of contractile proteins in skeletal muscle and how these are altered by aging, diet and exercise. Finally Dr. Amanda Wright's NSERC funded research seeks to better understand the fundamental role of lipid molecular structure and physical properties on pre-absorptive digestive events that regulate nutritional functionality. Collectively, the research programs of the applicants of this proposal require the extensive analysis of changes in gene expression as it relates to multiple systems, including skeletal muscle, adipose tissue, liver and gut function and metabolism. Thus, the applicants are requesting funding to purchase a new RT-PCR machine for these measurements. The data generated from the ongoing studies in the applicants laboratories will increase our basic understanding of the mechanisms controlling skeletal muscle, adipose tissue, liver and gut function and metabolism under a variety of nutritional and physiological stresses.
