Netherton Syndrome (NS) is a life-threatening skin disease caused by mutations in the SPINK5 gene. These mutations inactivate an endogenous serine protease inhibitor, called LEKTI. In human skin LEKTI inhibits the activity of skin kallikreins (KLKs). K
