Fluorescent dyes that emit light upon binding to double stranded DNA are crucial in applications such as quantitative Polymerase Chain Reactions (qPCR) and High Resolution Melt (HRM) experiments, both important techniques in molecular biology. These techniques have found a wide range of applications in life science, forensics, human health and beyond. While the dyes that are currently in use fulfill the requirements for qPCR and HRM to various degrees, many are expensive or not efficient enough for the applications. This project will investigate alternative dyes based on 1,8-naphthalimide core structures. The 1,8-naphthalimide fluorescent dyes are usually straightforward to synthesize, and some analogs have unique fluorescent properties that are suitable for qPCR and HRM applications. This research will generate 1,8-naphthalimide dyes that are readily available, and potentially suitable for both qPCR and HRM applications.
