Canadian produces about 9 billion eggs per year and about 27% are processed at egg breaking which yield lowvalue eggshell as a by-product. The Canadian poultry industry produces about 1.5 tons eggshell membrane(ESM) waste annually. The discovery of novel value-added applications for eggshell waste would haveimportant financial and environmental benefits. Eggshell-derived collagen and collagen-derived peptides are anattractive replacement for bovine collagen due to the concern over bovine spongiform encephalopathy (BSE).Only a few applications have been reported, including matrices for adsorption of heavy metal ions, biologicaldressings for burns, substrates for cell culture and platforms for enzyme immobilization. Further studies arerequired to validate the biological activity of ESM peptides for specific cosmetic application. Hypothesis: ESMpeptides can be applied in cosmetics including skin grafts, stimulate the growth of human skin fibroblasts,moisture, whitening and reduce the production of inflammation in skin. The objectives: 1)To assess theproliferation of fibroblasts on the collagen gel by ESM peptides in vitro, 2) To evaluate fibroblasts migrationand enzyme activity (hyuaronase, tyrocinase, elastase etc) in vitro cell culture model. The main targets of thisproject are 1) enhance fibroblast proliferation, 2) anti-inflammation and 3) whitening. Fibroblast migration isimportant factor for maintaining skill elasticity, moisture and we measure the effect of ESM peptides on thecell proliferation and hyuaronase/estase activity. Anti-inflammatory activity will be measured by testing TNF-?,MCP-1, IL-6 which are typical inflammatory mediators in human skill and they are related to anti-aging ofskin. Lastly, we will measure whitening activity of ESM peptides by means of testing tyrocinase activity andanti-UV effect. The ultimate goal of the project is to develop valued added ESM peptides from industrial wasteESM. ESM derived biomaterials are attractive to world markets where individuals have religious concernsregarding porcine or bovine sourced material.
