英文摘要
Significant developments made in the fields of nucleic acid chemistry and biology have solidified nucleic acid therapeutics' presence at the forefront of modern medicine.  Chemically synthesized oligonucleotides are now recognized as the third major drug discovery platform, in addition to small molecules and antibodies. Additionally, synthetic DNA and RNA provide valuable probes to study key structure -function relationships that offer us molecular -level insights of important biological mechanisms and processes. The automated DNA/RNA synthesizer (or "gene machine") requested in this application is the worldwide gold standard for producing multiple DNA and RNA strands simultaneously and at a scale that permits systematic evaluation of their three- dimensional structure and therapeutic activity. The instrument we have identified is unique because it allows for researchers to synthesize multiple strands independently, while incorporating up to 20 DNA/RNA modifications that will help us screen for the candidates with the most desirable properties for our applications in therapeutics and structural analysis. Students and researchers trained with this instrument will gain new insights into DNA/RNA structure and biology, and gain expertise in synthesis, analytical skills, and strategies to develop candidate molecules into useful drugs. This training is valuable to employers worldwide in the areas of chemistry and medicinal chemistry, especially as the field of DNA/RNA therapeutics continues to expand.
