软骨组织移植后期的存活一直是临床面临的问题。课题组前期利用颗粒软骨构建出条块状及外耳廓软骨移植体,具备良好的柔韧度和力学强度,但其缺乏真正的血供来源,存在不可控的吸收问题。研究证实软骨膜血管新生对软骨生长发育与再生起重要的调节作用。我们在预实验中发现保留完整兔耳软骨膜进行自体颗粒软骨原位回植,颗粒软骨能更好的存活。因此推测"调控移植床血管新生可改善自体颗粒软骨移植体的存活微环境"。为证明这种假说,本项目拟首先体外证实自体软骨膜和所构建的外源性软骨膜类似物(SVF细胞膜片、ADSC细胞膜片、脱细胞羊膜、VEGF基因修饰PLGA膜)促血管新生和软骨细胞存活的作用,再体内检测它们重建移植床血管新生对颗粒软骨移植体存活微环境的影响,然后探讨血管生成细胞/分子的时空效应在自体软骨膜和外源性软骨膜类似物重建稳定微血管网络中的调控作用,为改善颗粒软骨或软骨组织移植后存活与再生提供新策略和新思路。
英文摘要
Survival in the later stage of cartilage transplantation has been a clinical problem. In the previous study, we used diced cartilage to construct bar mass and external auricle cartilage implants, but it lacks real blood source and has the problem of uncontrollable absorption. Studies have confirmed that perichondrium angiogenesis plays an important role in regulating cartilage growth and regeneration. We found in the pre-experiment that the intact rabbit ear perichondrium was retained for autologous diced cartilage in situ replantation. The diced cartilage could better survival. Therefore, it is speculated that regulating angiogenesis in transplant bed can improve the survival microenvironment of autologous diced cartilage grafts. To demonstrate this hypothesis, the project intends to confirm the effects of the autogenous and exogenous perichondral analogues(SVF cell sheet, ADSC cell sheet, decellular amniotic membrane, VEGF gene-modified PLGA membrane) on angiogenesis and chondrocyte survival in vitro firstly. And observe the effects of remodeling angiogenesis in transplant bed by autogenous and exogenous perichondral analogues on the survival microenvironment of diced cartilage grafts in vivo secondly. Then to investigate the spatiotemporal effect of angiogenic cells/molecules in the reconstruction of stable microvessel network by autologous perichondrium and exogenous perichondral analogues. This study is to provide new strategies and ideas for improving survival and regeneration of diced cartilage or cartilage tissue after transplantation.
