英文摘要
Fuzhuan brick tea polysaccharides (FBTPS) could modulate the host lipid metabolism. The previous work showed the regulation of host lipid metabolism by FBTPS may be related to the modulation of Bacteroides plebeius in gut microbiota. However, the mechanism of interaction between FBTPS and B. plebeius is still unclear. FBTPS-2-1 is the main fraction in FBTPS. In this project, the interaction relationship between FBTPS-2-1 and B. plebeius is evaluated by model of anaerobic fermentation of single strain as well as metabolomics and transcriptomics. The potential polysaccharide Utilization Loci (PUL) for hydrolysis of FBTPS-2-1 was predicted and excavated by metagenome sequencing and transcriptomics of B. plebeius. The potential carbohydrate active enzymes (CAZymes) were expressed by Escherichia coli. The metabolism of FBTPS-2-1 by CAZymes was confirmed by evaluating enzymatic characterization of CAZymes. This project study the interaction of FBTPS-2-1 with B. plebeius based on the excavation of key PULs and CAZymes, which may not only clarify the possible mechanism of FBTPS in regulating lipid metabolism, but also provide theoretical support for the production and high-value utilization of Fuzhuan brick tea.
