如何通过提高水产动物HUFA的合成能力来降低其对鱼油的依赖是近年来水产动物营养研究的热点。课题组前期研究发现,拟穴青蟹体内HUFA合成能力可能缺乏或极低,但其肌肉组织具有较强的HUFA沉积能力,且HUFA沉积存在明显的组织选择特异性并随生长阶段的变换而呈现动态变化特点,推测沉积的HUFA主要来源于外源HUFA的高效摄入和转运。研究表明90%以上的长链脂肪酸吸收是由转运蛋白介导的。基于这些研究基础,本项目拟在探明青蟹对功能性HUFA的吸收速率、组织偏好性等特性基础上,在体外筛选偏好于转运HUFA的转运蛋白,然后结合体内体外实验系统分析该转运蛋白表达模式与不同生长阶段青蟹组织HUFA选择性沉积的动态变化规律间的关联性及其对外源HUFA的响应情况,最终阐明青蟹HUFA选择性沉积的可能机制,旨在独辟蹊径,为从提高机体对外源HUFA吸收沉积能力方面探索提高机体HUFA含量的新思路新途径提供理论依据。

In recent years, it is the focus of aquatic animal nutrition to reduce the dependence on fish oil by improving the capacity of HUFA biosynthesis in aquatic animals. Our previous studies found that mud crab, Scylla paramamosain might have no or very low capacity of HUFA biosynthesis. However, a strong ability of HUFA deposition was found in muscle tissue of mud crab, and there is an obvious tissue selection specificity for the HUFA deposition which varies dynamically with the change of growth phase. Therefore, we speculated that the high efficient intake and transport of exogenous HUFA may be the main source of the HUFA deposition in tissue. It was demonstrated that more than 90% of long-chain fatty acid absorption is mediated by transporters. Based on the results of these researches, this project is conducted to elucidate finally the possible mechanism of HUFA selective deposition in mud crab through analysis of the correlation between the expression patter of important transporters and the dynamic changes of HUFA selective deposition in different growth stages and the responses to exogenous HUFA after the characteristics of HUFA selective deposition were investigated, such as the absorption rate, tissue preference of functional HUFA and so on. This may provide theoretical basis for exploring new ideas and ways to improve the content of HUFA by increasing the ability of absorbing and depositing exogenous HUFA in aquatic animals.
