病毒感染介导心肌细胞损伤是导致病毒性心肌炎(VMC)的核心机制之一。我们前期研究发现病毒感染可通过calpain介导致线粒体结构功能异常,详尽机制不清。近期我们在VMC模型预试验中发现线粒体calpain异常活化可切割ATP合酶-α(ATP5A1)介导线粒体功能障碍;同时也观察到病毒感染引起线粒体功能障碍可激活NOD样受体家族蛋白3(NLRP3)炎症小体加重心肌损伤。综合上述工作我们提出假说:病毒感染激活线粒体calpain切割APT5A1致线粒体功能障碍并激活NLRP3炎症小体进一步加重心肌细胞损伤。本项目拟利用心肌线粒体靶向过表达calpain抑制剂calpastatin转基因小鼠及心肌特异性calpain亚基Capn-4敲除小鼠从表达与功能两个层面,采用Western blot、ELISA、免疫组织病理、透射电镜、线粒体炫观察等技术手段验证上述假说、阐明具体机制,以明确其科学价值。

Viral infection-mediated cardiomyocytes damage is one of the core mechanisms leading to viral myocarditis (VMC). Our previous researches revealed that viral infection caused cardiomyocytes damage through calpain activation. Recently, we found that virus infection could lead to abnormalities of both mitochondrial structure and function with calpain translocation to mitochondria, suggesting that calpain may participate in the pathogenesis of VMC by regulating mitochondrial function. It has been reported that calpain abnormal activation could result in mitochondrial dysfunction by cleaving ATP synthase-α (ATP5A1) in the septic cardiomyopathy model. Meanwhile, we found preliminarily that mitochondrial dysfunction mediated by virus infection could activate NLRP3 inflammasome and aggravate myocardial injury. Therefore, we propose the hypothesis that pathologic calpain activation caused by virus infection contributes to mitochondrial dysfunction by cleaving ATP5A1 and activating NLRP3 inflammasome which further aggravate myocardial injury. In this project, we intend to verify the above hypotheses and clarify the mechanisms from both expression and function perspectives by using myocardial-specific mitochondrial overexpressing calpastatin, the endogenous inhibitor of calpain, transgenic mice, and myocardial-specific calpain knockout mice and utilizing western bolt, ELISA, immunohistopathology, transmission electron microscopy and mitochondrial flash obseration to clarify the roles of calpain in VMC pathogenesis.
线粒体代谢异常造成的心肌细胞损伤是心肌炎发生发展的重要病理过程。我们既往发现,在CVB3导致的病毒性心肌炎中calpain活性增高且可促进CVB3病毒复制。Calpain被过度激活会通过限制酶切作用水解多种蛋白,造成组织损伤,而calpain的活化又与线粒体动力学失衡有关。为了深入探讨其可能机制,本项目利用心肌线粒体靶向过表达calpain内源性抑制剂calpastatin的转基因小鼠(Tg-mtCAST)建立病毒性心肌炎模型,同时在体外利用线粒体特异性CAST过表达腺病毒 (adeno-mtCAST) 感染心肌细胞,分别通过抑制calpain活性以及下调calpain表达两种方式,明确病毒感染通过线粒体calpain介导的心肌线粒体损伤的科学事实并阐明具体机制。. 研究发现,在CVB3感染的心肌炎中calpain活性增加、NLRP3炎症小体激活、线粒体结构功能障碍。体内分别抑制calpain活性、抑制NLRP3炎症小体活性、以及通过清除线粒体ROS改善线粒体功能,都能够阻止CVB3感染导致的心肌损伤,改善心功能。进一步的研究发现,抑制calpain活性能够遏制NLRP3炎症小体的激活、改善线粒体呼吸链功能、降低线粒体ROS的产生,而清除线粒体ROS能够抑制NLRP3炎症小体的激活,并且逆转calpain过表达对NLRP3炎症小体活性的促进作用。同时阐明了CVB3感染后,心肌细胞中calpain活性增强,通过损害线粒体的功能,促进线粒体ROS过度产生,从而激活NLRP3炎症小体导致心肌损伤这一调节机制。更深层次的,我们的研究还证明了CVB3刺激下,Calpain通过转移至线粒体内,直接切割ATP5A1,引起其功能失常,ATP合成障碍,从而过度的ROS产生。本研究较全面地从动物、细胞水平阐明了CVB3心肌炎中calpain作用的新机制,为后续治疗靶点研究提供了思路。
