雄性不育材料在甘蓝杂种优势利用中发挥着重要作用,同时也是研究植物生殖发育调控的重要资源。本项目组在前期发现了一份受隐性单基因控制的雄性不育突变体83121A,研究发现花粉外壁缺失是导致其小孢子败育的主要原因,并通过功能互补验证确定了该雄性不育是由BoCYP704B1突变所致。但BoCYP704B1在甘蓝花粉外壁发育中的作用及其上游调控机制尚不清楚,本项目针对以上问题,重点开展以下三方面研究:(1)通过基因编辑技术进一步分析BoCYP704B1的功能;(2)利用生物信息学、酵母异源表达及酶学分析等方法,解析BoCYP704B1的生化特征和催化活性;(3)通过启动子表达分析和酵母单杂交技术挖掘BoCYP704B1的上游转录调控因子。预期研究结果对于解析BoCYP704B1在甘蓝花粉外壁发育中的生物学功能、丰富和完善甘蓝花粉发育调控机制具有重要意义。

Male sterile mutants play an important role in the utilization of heterosis in cabbage, and are also important materials to study the development and regulation of plant reproduction. 83121A is a spontaneous male sterile mutant identified from cabbage. Genetic analysis indicated that the male sterility is controlled by a single recessive gene. Pollen wall formation in the 83121A mutant was severely defective, with a lack of exine. Complementation test determined that the mutation of BoCYP704B1 is the cause of the male sterility of 83121A. However, the role of BoCYP704B1 in the development of exine and its upstream regulation mechanism are not clear. In view of the above problems, this study will carry out the following research. (1) Functional analysis of BoCYP704B1 will be conducted by gene knockout. (2) Biochemical characteristics and catalytic activity of BoCYP704B1 will be studied by bioinformatics analysis, heterologous expression and enzymatic analysis. (3) The upstream regulation genes of BnCYP704B1 will be analyzed by promoter truncation experiment and yeast one-hybrid techniques. The implement of this project will deepen our understanding of the function of BoCYP704B1 in the exine development and it will be of great significance to enrich and perfect the regulation mechanism of pollen development.
