近红外二b区(NIR-IIb, 1500-1700nm)荧光成像是生物医学成像领域的研究热点。然而,NIR-II光激发的NIR-IIb荧光成像技术在前列腺癌靶向显像及光热治疗研究尚未见报道。本项目拟通过:合成前列腺癌特异性标志物PSMA的单克隆抗体(anti-PSMA mAb);合成具有出色NIR-IIb荧光特性和光热性能的新型半导体聚合物P16并偶联anti-PSMA mAb制备P16-anti-PSMA mAb纳米粒子(P16-aPSMA NPs);搭建NIR-IIb荧光活体和显微成像系统,利用前列腺癌PDX模型,以P16-aPSMA NPs为NIR-IIb荧光/光热多功能探针进行前列腺癌靶向显像和NIR-IIb荧光成像引导下特异性光热治疗研究。本项目研究NIR-II激发的NIR-IIb荧光成像技术应用于前列腺癌靶向显像和特异性光热治疗的可行性,为前列腺癌的精准诊断及治疗提供新的思路。

Near-infrared IIb (NIR-IIb, 1500-1700 nm) fluorescence imaging is becoming a research hotspot in the bioimaging field. However, the study of NIR-II excitable NIR-IIb fluorescence imaging in targeted imaging and photothermal therapy for prostate cancer have not been reported yet. This project intends to: Synthesize the monoclonal antibody of prostate cancer specific marker PSMA (anti-PSMA mAb); Prepare the prostate cancer targeting P16-anti-PSMA mAb nanoparticles (P16-aPSMA NPs) by coupling anti-PSMA mAb to the surface of the newly synthesized semiconductor polymer P16 which possess strong NIR-IIb fluorescence characteristics and excellent photothermal properties under NIR-II light excitation; Set up NIR IIb fluorescence whole-body imaging and microimaging system; Establish prostate cancer PDX model; Achieve prostate cancer targeted NIR-IIb fluorescence imaging and NIR-IIb fluorescence imaging guided specific photothermal therapy of prostate cancer in vivo and in vitro utilizing the P16-aPSMA NPs as NIR-IIb fluorescence / photothermal multi-functional probe. This project aims to explore the feasibility of NIR-II excitable NIR-IIb fluorescence imaging technology in targeted imaging and spesific photothermal therapy of prostate cancer, and provide a new idea for precise theranostics of prostate cancer.
