肺癌是我国发病率和死亡率第一的恶性肿瘤,发展针对肺癌的分子影像方法符合国家“提高人口质量和全民健康水平”的战略目标。本课题组已应用超极化技术成功点亮了磁共振肺部“盲区”,在国内首次实现了信号增强倍数达到55000倍的人体肺部超灵敏磁共振成像(129Xe MRI)。但是,129Xe作为惰性气体没有靶向性,难以用于分子影像研究。本项目拟以两亲性V型多芳香骨架作为基础单元,以肺癌细胞靶向性分子对基础单元进行修饰,进而自组装成可装载129Xe的“肺癌靶向性芳香族胶束分子笼”,即能用于肺癌129Xe MR分子影像研究。此外,“芳香族胶束”还具有自发荧光,其“分子笼”空腔还能运载其他造影剂进行1H MRI、荧光成像等,以成熟成像手段验证129Xe MRI这种新方法的可靠性,探索将129Xe MR分子影像传感应用于肺癌早期超灵敏诊断的潜在可能性。
英文摘要
As a malignant tumor with the higest morbidity and mortality in China, lung cancer has been given rising attentions along with environmental deterioration. Development of new molecular image techniques for targeted determination of lung cancer is a major demand to national heath. It is hard to image the lung by clinical used MRI, because of the little density of H in lung cavity structure. This problem has been solved by Hypersensitive 129Xe MR, which increases MR sensitivity 55,000 times. 129 Xe MRI provides a new imaging technique for lung structure and function diagnosis with non-radiation and non-invasion. However, 129Xe MRI has no targeting due to the chemical inertness of Xe. In this project the amphiphilic V-type polyaromatic skeleton will be used as the basic unit, and the lung cancer cell targeting molecules has been chosen for basic unit modification. Then, a 129Xe-loaded lung cancer targeting aromatic micelle molecular cage will be self-assembled for lung cancer 129Xe MR molecular imaging. Furthermore, the aromatic micelle molecular also can be used in 1H MRI and fluorescence imaging, evaluate the newly developed 129Xe MRI by clinically used imaging techniques, and provide a new tool for molecular imaging and cancer treatment.
