花青素使菜豆豆荚表现出紫色,这不仅可使菜豆豆荚表现出更具吸引力的紫晕或紫色条纹,并且对植物应对环境胁迫起到重要保护作用,因此对菜豆中花青素合成相关基因的研究有助于今后菜豆的育种改良。申请人前期以菜豆‘A18-1’种子为试材,采用钴-60辐射诱变处理的方法,创制出1份紫化突变体pv-pur,其胚轴、茎、叶脉、花、豆荚均表现出紫色,其它性状与野生型基本一致。遗传分析证明了pv-pur的紫化性状为单基因显性遗传,遂将突变基因命名为PV-PUR。利用BSA混池测序的方法,已将PV-PUR定位于Chr06染色体3740000-9590000范围内。本项目拟进一步开展PV-PUR基因克隆与鉴定工作,同时探索紫化突变基因引起突变体抗逆性上的变异。研究成果将为揭示紫化突变基因的功能与作用,更好的利用这一突变资源奠定基础。

The anthocyanins that make the bean pods show purple can not only make the bean pods show more attractive purple halo or purple stripe, but also play an important role in protecting plants from environmental stress. Therefore, the research on the genes related to the anthocyanin synthesis in snap bean will be helpful to the breeding and improvement of snap bean in the future. In previous research, the seeds of snap bean ‘A18-1’was employed as the plant material, and a purple mutant pv-pur was created by the method of 60Co-γ radiation mutation. Its hypocotyl, stem, vein, flower and pod all showed purple color, and other characters were basically the same as the wild type. Genetic analysis indicated that the purple trait of pv-pur was controlled by a simple dominant nuclear gene, designated as PV-PUR. Using BSA mixed cell sequencing technology, PV-PUR was located in the range of Chr06 chromosome 3740000-9590000. In this project, the study on cloning and functional identification of PV-PUR will be performed. At the same time, the mutation of stress resistance in pv-pur which caused by purple mutation gene will be explored. The research results will lay the foundation for revealing the function purple mutation gene and making better use of this mutation resource.
