肠道菌群与宿主的成瘾行为及大脑之间存在着密切的联系,其中肠道微生物的扰动对神经系统的调节发挥着关键作用,通过改变肠道菌群治疗成瘾成为一种新思路。针灸可以通过作用于不同穴位改善失调菌群的比例,对肠道菌群具有调节作用。基于此,本课题建立氯胺酮成瘾恒河猴模型并进行穴位电针治疗,采用BOLD-fMRI技术观察电针对氯胺酮成瘾恒河猴脑功能活动变化的影响,明确电针治疗对成瘾相关脑区功能活动的调节在改善成瘾戒断症状中的关键作用;通过ERIC-PCR指纹图谱分析、免疫组织化学法、半定量RT-PCR、Western blotting蛋白印迹法等方法,观察电针对氯胺酮成瘾恒河猴脑区及胃肠道不同部位相关蛋白及其mRNA表达及肠道菌群指标变化的影响,证实电针治疗氯胺酮成瘾可能是通过对菌-肠-脑轴不同靶点肠道菌群及相关脑肠肽的调整,从而实现对相关神经元的保护作用,为临床运用电针和调节肠道菌群防治成瘾提供实验依据。

Gut bacteria is closely related to the host's addictive behavior and brain, and it was found that the intestinal microbial disturbance on the regulation of the nervous system plays the most crucial role,it has become a new way to treat addiction by changing gut bacteria. Acupuncture can improve the proportion of dysregulated flora by acting on different acupuncture points, and has a regulating effect on gut bacteria. Based on this idea, in our project the model of ketamine addiction in rhesus monkeys will be established by the conditioned response induced by bell stimulation, and the electroacupuncture therapy will be carried out, the BOLD-fMRI technique is used to observe the effect of electroacupuncture for changes in functional brain activity of ketamine addiction rhesus monkey, revealing that electroacupuncture therapy plays a key role in improving the withdrawal symptoms of ketamine addiction by regulating the functional activities of the brain areas related to addiction; Taking the ERIC-PCR fingerprinting technique, immunohistochemistry, RT-PCR, and Western blotting analysis as the detection method, to observe the effects of electroacupuncture on the expression of related proteins and their mRNA in different parts of brain and gastrointestinal tract, and also the changes of gut bacteria indicators of rhesus monkey with ketamine addiction, thus to prove that the protective effects of ketamine addiction related brain neurons and enteric neurons induced by electroacupuncture are realized by regulating the gut bacteria and brain-gut peptide in different targets during the bacteria-gut-brain axis, and further more, it will provide powerful evidence for clinical application of electroacupuncture and regulation of gut bacteria to prevent and treat addiction.
