对虾是我国海水养殖的主要品种之一,近年来种质退化、环境恶化和病害频发等问题制约了对虾产业的健康持续发展。而解决问题的关键之一是解析调控对虾重要性状的基因功能,从而深入理解对虾这一物种。我们近期研究发现对虾血清中存在一种高度保守的细胞因子-类神经营养因子MANF,其敲降可导致对虾炎症反应明显减弱,提示对虾MANF可能与炎症反应的调控有关。本项目拟在此基础上,首先通过体内敲降和体外重组蛋白处理原代血淋巴细胞技术对该蛋白的胞内和胞外功能进行分类研究;其次通过pull down和免疫沉淀技术筛选和验证其互作蛋白,并结合筛选到的互作蛋白对其胞内和胞外炎症调控功能进行分类机制探索;最后通过攻毒实验分析该蛋白在对虾抗病过程中的功能。预期研究成果将为对虾高产抗逆育种提供新的分子标记物,为对虾的免疫学研究做出贡献。

Shrimp is one of the major mariculture species in China. In recent years, problems such as germplasm degradation, environmental deterioration and frequent diseases restrict the health and sustainable development of shrimp industry. To solve these problems, one of the keys is to analyze the function of the gene that regulates the important traits of shrimp, so we can understand shrimp in depth. In recent studies, we have found that there is a highly conserved cytokine MANF in shrimp serum, and its knockdown can significantly reduce shrimp inflammation, suggesting that this gene is highly likely related with inflammation regulation. On this basis, the project, firstly, studies the intracellular and extracellular function of this protein by knockdown in vivo and in vitro recombinant protein treatment of primary hemolymph cells; secondly, screens and validates its interacting proteins by pull-down and immunoprecipitation techniques, and explores its intracellular and extracellular inflammation regulation mechanisms via the screened interacting proteins; finally, analyzes the function of this protein during shrimp anti-pathogen process by challenge assay. The expected results will provide new markers for shrimp high yield and stress resistance breeding, and contribute to shrimp immunological research.
