大豆是我国饮食结构中蛋白和油分的主要来源之一,如何实现大豆品种蛋白质含量与油分含量的双高是育种和生产加工企业的需要。本研究以一套野生大豆全基因组导入系群体为基础,基于多年的表型筛选,获得蛋白质含量和油分含量组合有明显差异的4份材料,在种子发育重要时期(早熟期,中熟期和种子成熟期)取样进行转录组测序,鉴定不同时期不同材料之间的蛋白油分含量差异表达模式。同时,整合国内外近30年发表的大豆蛋白质和油分含量QTL信息进行Meta分析。将Meta-QTL结果与转录组数据联合分析,初步构建了蛋白质和油分含量协同调控网络。本项目将针对筛选出的特殊材料进一步构建剩余杂合体群体,完成QTL的精细定位,同时结合已构建的调控网络交互验证候选基因,并对关键基因进行大豆转基因功能验证,开发标记在次级群体验证和等位分型分析。研究结果可为大豆的高品质育种提供理论依据和分子育种指导。

Soybean is a main source of protein and oil for people consumption in China. How to achieve the double height of soybean protein content and oil content is the requirement of breeder and producer. Based a set of chromosome segment substitution lines (CSSLs), four CSSLs were selected with different seed oil and protein contents to characterize their three years of phenotype selection in the field. Using strand-specific RNA-sequencing analysis, we profile the time-course transcrotpome patterns of soybean seeds at early maturity, middle maturity and dry seed stages in different materials. Meta –analysis was also conducted to integrate recent 30 years soybean protein and oil content QTLs. Meta-QTL and transcrotpome data were analyzed together to construct the co-expression networks. In this study, residual heterozygous lines (RHLs) were selected based on the introduced fragment of four seleted CSSLs, and QTL fine mapping was conducted for these RHLs, furthermore, the candidate genes were verified by combining the established regulatory networks. The hub genes were verified by soybean transgenic research, and the markers of hub genes were developed in sub-population verification and allelic analysis based on resequencing. The research results can provide theoretical basis and molecular breeding guidance for high quality breeding of soybean.
