桑寄生是我国大宗而又繁殖困难的寄生药材,而鸟类传播是其主要繁殖途径。我们前期研究表明鸟类摄食对桑寄生种子萌发有促进作用,已有研究也证实鸟类摄食可促进种子萌发,但主要是侧重研究鸟类传播与植物种子扩散的关系,而鸟类摄食对种子萌发的具体作用机制研究鲜有报道。基于鸟消化系统的复杂性,种子经过鸟消化后是否携带了可促进萌发的有益真菌?其生理生化如何变化?是否启动了促进萌发的基因与蛋白?这些都值得去探讨。因此,本研究围绕新鲜种子经鸟摄食消化后到萌发的阶段:①分析鸟摄食与种子萌发的相关性;②分离、筛选并鉴定促进种子萌发的有益真菌;③分析种子生理生化的变化;④采用RNA-seq技术和iTRAQ技术进行转绿组和蛋白质组研究,揭示种子内部基因与蛋白的表达规律。从微生物、生理生化、转录组、蛋白组等综合阐明鸟摄食对桑寄生种子萌发的作用机制。不仅为桑寄生繁殖提供理论依据,也为鸟类传播与植物种子扩散关系的研究提供参考。
英文摘要
Taxillus chinensis (DC.)Danser is one of the most commonly used traditional chinese medicines, which is parasitic but difficult to reproduce plant, and the main reproductive pathway depends on the bird transmission. Our previous studies have shown that the seed germination of T. chinensis increased through the bird ingestion. The previous studies have also confirmed that the promotion role of the bird ingestion on seed germination, but which all emphasized the synergistic effect relationship between the diffusion of plant seeds and the bird transmission. However, there are few researches on the mechanism of bird ingestion on plant seed germination. Because of the complexity of the bird's digestive system, there are many scientific questions worth discussing about the effects of the bird ingestion on plant seed germination, for example, whether the surface of seeds by the bird digestion carry the beneficial fungi that can promote seed germination? How does the physiological- biochemical factors in the seeds change? Whether the genes and proteins of promotion germination in the seeds are activated? Therefore, we will conduct the following researches about the effect of the seed germination of T. chinensis after ingested by birds: ① Analysis of correlation between bird ingestion and seed germination; ② Isolation, screening and identification of beneficial fungi promoting seed germination; ③Analysis the physiological-biochemical changes of seeds; ④ Study of the transcriptome and troteome by RNA-seq and iTRAQ techniques for revealing the expression of genes and proteins in the seeds. Such information is very important in elucidating the mechanism of seed germination of T. chinensis after ingested by birds from microorganism, physiology-biochemistry, transcriptome, proteome and so on. It not only provides a theoretical basis for the reproduction of T. chinensis, but also provides a reference for the research on the relationship between bird transmission and plant seed diffusion.
