Electroacupuncture (EA) can improve the dysfunction of hypothalamus-pituitary -adrenal axis caused by surgical trauma, while its neurobiological mechanisms remain to be clarified. In this study, we will focus on the newly discovered key molecule secretagogin (SCGN), the trigger of stress process, which is the key molecule regulates the release of CRH. We will explore the role of SCGN in hypothalamic PVN in EA modulating the abnormal function of HPA axis induced by acute surgical trauma, and further elaborate the mechanism of EA. It has been studied for decades on hypothalamic NMDA receptor regulation of CRF synthesis and release, whether hypothalamic NMDA receptors and its activated intracellular signaling pathways participate in the EA regulation of HPA axis dysfunction in rats during perioperative period is unclear. Whether they participate in EA regulated the dysfunction of HPA axis during perioperative period through affecting the synthesis and expression of SCGN has not been reported. Whether hypothalamic NMDA receptor-related microRNAs participate in the EA regulation of HPA axis dysfunction in rats during perioperative period deserves further study. On the basis of the previous studies, we will explore the mechanism of SCGN, NMDA receptors and their key molecules in intracellular signaling pathway, and microRNAs targeted NMDA receptor participating in the EA regulation of HPA axis dysfunction in rats from cellular and molecular levels on partial hepatectomy model, so as to provide experimental and theoretical basis on the EA alleviating stress response during perioperation period, and provide a theoretical basis on EA popularization in clinical practice.
