Dental implantology attracts attention as an important treatment modality for edentulism because of its excellent predictability. To achieve the stability of implant in the bone, 3 to 6months are required for the bone healing. If this duration can be truncated, it may be beneficial for both doctors and patients. Recently, immediate loading procedure has been introduced into the implantology, however, there are few scientific evidences for such treatment.In the present study, we hypothesized that immediate loading is safely performed, without any complication, and we intended to elucidate the relationship between bone healing and the timing and quantity of the loading. In 2004, we examined the appropriate method for the loading and how the force spread into the surrounding bone using finite element method. Based upon these results, we obtained several numbers of canines and teeth extraction was performed. In 2005, we performed the histological analysis around the loaded dental implants and then we analyzed the protein expression pattern around the bone adjacent to the loaded implant, using microarray method. Two implants were installed into canine mandible, and then static load was operated between implants using rapid expansion screw for orthodontics. Seven days after the load, the bone around the implant was disrupted without the presence of osteoclasts. In microarray analyses, some kinds of proteins revealed the alteration of mRNA expression pattern in response to the loading. It was suggested that the alteration of the protein expression might play a role in the disturbance of osseointegration.
