Physiologisch veränderte Bedingungen können die Differenzierung der asexuellen und sexuellen Fruchtkörper (Konidiophoren bzw. Cleistothecien) des filamentösen Pilzes Asperigillus nidulans beeinflussen. Aminosäuremangel bewirkt eine definierte, reversible Blockierung der sexuellen Fruchtkörperbildung auf einem intermediären Diffenrezierungsstadium (Mikro-Cleistothecien). Die Überexpression des Histidin-Biosynthese-Gens hisHF führt zur Ausbildung morphologisch stark veränderter Sporenträger für die asexuellen Konidien. Diese Quervernetzung zwischen Metabolismus und Entwicklungsabläufen ist bisher kaum untersucht und soll aufgeklärt werden. Metabolische Signale wirken auf das genetische Netzwerk der "Cross-pathway-Control" der Aminosäurebiosynthese. Wir werden prüfen, wie Veränderungen in der Konzentration vorhandener Aminosäurebausteine Differenzierungsprogramme beeinflussen können. Ausgangspunkt der Untersuchungen sind die regulatorischen Gene cpcA und cpcB sowie die von ihren Genprodukten regulierten Ziele, insbesondere die Gene für die Histidin-Biosynthese.
