英文摘要
GTP-Cyclohydrolase II katalysiert die Abspaltung von Pyrophosphat und Formiat aus GTP. Das resultierende Triaminopyrimidinderivat entsteht als erstes Intermediat der Vitamin B2-Biosynthese. Da zahlreiche humanpathogene Mikroorganismen Flavine nicht aus der Umgebung aufnehmen können, sind sie zwingend auf die Eigensynthese von Vitamin B2 angewiesen. Daher ist das Enzym GTP Cyclohydrolase II ein potentieller Angriffspunkt für antibakterielle bzw. antimykotische Wirkstoffe. In Zusammenarbeit mit Kristallographen sollen Strukturen von GTP Cyclohydrolase II aus apathogenen sowie aus pathogenen Mikroorganismen und aus Pflanzen aufgeklärt werden. Der Reaktionsmechanismus soll untersucht werden durch gemeinsamen Einsatz von ortsspezifischer Mutagenese, Enzymkinetik und spektroskopischer Methoden. Die Arbeiten sollen u.a. Voraussetzungen für die Entwicklung neuartiger Antiinfektiva bzw. Herbizide schaffen.
