The advent of new biomedical imaging technique has enabled significant progresses in how we image humans and animals. In particular, the emergence in the mainstream of new modalities, such as diffuse optical imaging, has enabled researchers to image hemodynamics in vivo in a non invasive fashion. In parallel, associated with the emergence of endogenous diffuse optical imaging in humans, a new approach is emerging enabling to follow molecular products injected in animals, in vivo, through fluorescence. This new imaging technique, referred to as molecular imaging holds great promises: whereas one would detect diseases through anatomical changes and anomalies, we now can use new methods to provide a minimally invasive characterization and measurement of biological processes at the cellular and molecular levels of living organisms.Up until now, most optical imagers have been constrained by diffusion and heterogeneity. The injection and detection of infrared light in tissues (with and without fluorescence) is subject to the complex variations of optical properties of the medium. For many applications, in particular oncology, it is sometimes sufficient to ignore this, but as research advances, researchers are becoming more acutely aware of the limitations of this method, in particular for spatial resolution and quantification.The aim of this proposal is to develop techniques to address these limitations. We propose to address these limitations in two ways: first by developping a multi-spectral fluorescent imager providing more information about tissue optical properties. Second to create a dictionnary of template mice models from another modality (MRI) that can be used as prior in the reconstruction. The pre- computation of Green's functions on this template will be used to perform image reconstruction.The developments originating from this proposal will benefit the partner company by increasing its product line quantification ability.
