CD23, a type II transmembrane glycoprotein, belongs to the C-type (Ca2þ-dependent) lectin-like superfamily. The CD23 protein is cleaved from cell surfaces as a soluble form and exerts pleiotropic activities in addition to the regulation of IgE responses and B-cell-activation and-growth [1]. CD23 is widely distributed in many normal cells, including T and B lymphocytes, monocytes, and follicular dendritic cells, and its expression is subject to different regulations by various stimuli [2, 3]. CD23 expression is a useful and pertinent diagnostic marker to differentiate between chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL), mantle cell lymphoma (MCL), and other indolent lymphomas, and has been linked to their clinicopathological findings and prognoses [4, 5]. Thus, the precise evaluation and analysis of CD23 expression in target cells by flow-cytometric immunophenotyping (FCIP) and immunohistochemistry (IHC) are the key and critical processes for the strict diagnosis and its relevant most appropriate therapeutic choice [6, 7]. The interaction of CD23 with IgE is also a therapeutic targeting mechanism to prevent allergic diseases [8].In the clinical setting, the surface CD23 expression is evaluated in anticoagulated whole peripheral blood, bone marrow aspirates or single cell suspensions from their relevant flesh tissues, by the direct immunofluorescence staining FCIP analysis [9-12]. Therefore, primary antibodies against CD23 may be directly and simultaneously incubated with target cells in reaction solutions that contain various anticoagulants, including ethylenediaminetetraacetic acid (EDTA-2Na), heparin or acid citrate dextrose.
