Oxidative stress and redox regulation play a key role in development and hyphal growth in Aspergillus nidulans. The discovery of the central CCAAT-binding complex (AnCF) as a key player in redox regulation, growth and development allows to analyse at the molecular level how the reactive oxygen species (ROS)-signaling network controls apical growth and development of A. nidulans. This will be achieved by applying transcriptome and proteome analyses which will result in the identification of key genes / proteins regulated by both the NoxA (NADPH oxidase) and AnCF at different developmental stages like vegetative growth, asexual and sexual development. Among the already identified genes are genes most likely involved in cytoskeleton organisation like tubulin-alpha 2. These genes and further genes identified by functional genomics will be analysed, e.g., phenotypes of generated knock-out mutants under the different developmental conditions. Promoters of selected genes will be investigated for binding by AnCF, protein interaction partners by surface plasmon resonance analysis. NoxA dependent oxidation of HapC will be studied by bifunctional complementation assays using split GFP fused to AnCF subunits HapC and HapE. This study will contribute to identify the regulatory ROS-dependent network required for growth and development.
