Das bisher die tiefgehendsten Untersuchungen zu cis-/trans-Elementen von Sekundärstoffen in Pilzen erlaubende Modellsystem stellt die Penicillinbiosynthese in Aspergillus nidulans dar. Es wurden von uns mehrere trans-Faktoren isoliert: AnCF ist ein CCAAT-bindender Komplex, der aus den Proteinen HapB, HapC und HapE besteht. AnBH1 ist ein Transkriptionsfaktor, dessen Bindestelle mit einer der AnCF-Bindestellen überlappt. Ein weiterer, voraussichtlich mit AnCF interagierender trans-Faktor soll isoliert werden , um zu einem kompletten Bild der in die Biosynthese eines pilzlichen Sekundärstoffes involvierten cis/trans-Elemente zu gelangen. Da Arabidopsis thaliana im Gegensatz zu allen anderen bisher untersuchten Organismen 23 Hap-Homologe kodiert, soll untersucht werden, ob die pflanzlichen Hap-Proteine redundant in ihrer Funktion sind, oder ob nur bestimmte Hap-enthaltende Komplexe, falls überhaupt, in die Regulation von Sekundärstoffgenen involviert sind. Die Untersuchung der Regulation der trans-Elemente über posttranslationale Mechanismen soll in die Untersuchungen mit einbezogen werden. Mittels Proteom-Analysen von Deletions-Mutanten soll das regulatorische Netzwerk, in das die Biosynthese von Penicillin eingebunden ist, aufgeklärt werden. Dadurch hoffen wir, zu einem Modell der Evolution eines pilzlichen Sekundärstoff-Biosyntheseweges zu gelangen.
