Unsere Daten zeigen, dass Tumorzellen in Thrombozyten über eine Aktivierung und Sekretion der sauren Sphingomyelinase (Asm) Ceramid bilden und Integrine aktivieren. Hämatogene und lymphogene Tumormetastasierung ist in Asm-defizienten Mäusen um ca. 95% vermindert. Im vorliegenden Projekt untersuchen wir, über welche molekularen Mechanismen a) Melanomzellen die Asm in Plättchen aktivieren und die Sekretion des Enzyms vermitteln, b) sezernierte Asm Tumorzellen aktiviert und c) welche Rolle die Asm bei lymphogener Metastasierung von Melanomen spielt.
