Proapoptotische Bcl-2-ähnliche Proteine wie Bax. Bad oder Bak spielen bei sehr vielen Formen des programmierten zelltodes (Apoptose), z.B. nach Stimulation verschiedener pro-apoptotischer Rezeptoren, Behandlung mit ionisierenden Strahlen, UV-Licht, etc. eine entscheidende Rolle. Eine wichtige Funktion dieser Proteine wird zudem bei vielen degenerativen Erkrankungen, z.B. M. Parkinson, akuten Gewebsschädigungen, z.B. ischämischen Hirninfarkt, oder Krankheiten mit vermindertem Zelltod, z.B. Tumoren, vermutet. Trotz dieser überragenden Bedeutung pro-apoptotischer Bcl-2-ähnlicher Proteine bei der Regulation von Apoptose, sind die molekularen Mechanismen, über die diese proteine Apoptose induzieren, weitgehend unbekannt. Vorläufige Daten unserer Arbeitsgruppe zeigen, dass der Kaliumkanal Kv1.3 in Mitochondrien exprimiert ist, mit Bax interagiert und Bax-induzierte Apoptose vermittelt. Im vorliegenden Antrag sollen die molekularen Mechanismen, über die pro-apoptotische Bcl-2-ähnliche Proteine mit dem mitochondrialen Kaliumkanal Kv1.3 interagieren und Apoptose induzieren, charakterisiert werden.
