The influence of antigen distribution on quality and repertoire of CD8+ T cell responses

CD8+T淋巴细胞通过共刺激抗原提呈树突状细胞(DC)识别特定的多肽/MHC-I类复合体。此后，它们分化为效应性和记忆性细胞毒性T淋巴细胞(CTL)，能够提供对病原体感染的保护。由于其在T细胞“启动”中的中心作用，树突状细胞是抗体靶向的关键候选细胞，并被用作临床上的细胞疫苗。利用只有DC才能激活CD8+T细胞的转基因小鼠，我们将分析选择性DC激活对CD8+T细胞效应和记忆反应的定量和定性影响。

CD8+ T lymphocytes recognize specific peptide/MHC class I complexes in the context of costimuli on antigen-presenting dendritic cells (DCs). Thereafter they differentiate into effector and memory cytotoxic T lymphocytes (CTL) able to confer protection against pathogen infection. Due to their central role in T cell “priming”, DCs are key candidates for antibody targeting and are used as cellular vaccines for clinical purposes. Using transgenic mice where only DCs can prime CD8+ T cells, we will analyse the quantitative and qualitative influence of selective DC-priming on CD8+ T cell effector and memory responses.
