To date, most studies of bacterial small regulatory RNAs (sRNAs) have focused on the regulation of a single target mRNA. However, there has been increasing evidence that sRNAs might commonly act to regulate multiple targets. This project aims to fully understand the mechanisms and regulations by GcvB sRNA, for which a large number cellular targets have been predicted in Salmonella and other bacteria. GcvB is a widely conserved sRNAs that appears to globally regulate amino acid uptake and metabolism. There is evidence to suggest that, in order to regulate multiple targets, GcvB uses a variety of mechanisms, but these remain poorly understood. Our study of GcvB will help us better understand how sRNAs act as translational repressors, and which factors facilitate multiple target regulation. We will also determine how GcvB itself is regulated, and unravel the physiological meaning of GcvB-mediated regulation. In addition, we will endeavour to develop new methodology based on deep sequencing and FACS analysis that will be of use for other projects in SPP1258.
