I.Chloropeptinの絶対構造決定と、chloropeptinとcomplestatinの相互変換Chloropeptin(1)を6N塩酸で加水分解し、得られた4-hydroxyphenylglycine(2)および3,5-dichloro-4-hydroxyphenylglycine(3)の絶対配置を施光度、キラルTLCの結果からD体と決定した。残りのN-methyltyrosine(4)、3、4-dihydroxyphenylglycine(5)およびtryptophan(6)の絶対配置については、分子動力学法とNMRから推定した。可能性のある8つのジアステレオマ-を分子動力学計算で構築し、NMRから得られた二面角と距離情報を満足するジアステレオマ-を探した。その結果、4をL体、5、6をD体とした場合のみ、NMRの情報を全て満たした。また、1を6N塩酸-チオグリコール酸で加水分解し、得られた4をHPLCの結果からL体と決定した。この結果は分子動力学計算と一致したので、1の絶対構造を決定するに至った。1とcomplestatin(7)の相互変換は、これらのアルコール溶液の長期保存で確認された。また、メタノールートリフルオロ酢酸(1:1)溶液を乾固することにより7から1への変換が達成された。しかし、1から7への相互変換が認められなかったので、7から1への変換が容易であることが示唆された。II.Chloropeptinと逆転写酵素阻害剤の抗HIV活性に対する相乗効果最近、AIDSの治療法として作用の異なる薬剤の併用療法が注目されており、1と逆転写酵素阻害剤のAZT、ddI、ddCおよびnevirapineとの相乗効果を検討した。その結果、併用で相乗効果が認められたことから、ウイルスの細胞への接着・侵入阻害剤が併用療法に利用できる可能性を示した。
