セミノゲリン(SgII)の精子への結合性およびプロテインCインヒビター(PCI)との相互作用について解析した.また生理的役割を解析する目的で,健常人および男性不妊患者の精漿中SgIIとPCI濃度を測定し,病態との関連性について検討した.(1) 洗浄した精子とFITCを用いてラベル化したSgII(FITC-SgII)を反応させ,精子に対するSgIIの結合性を蛍光顕微鏡下に観察した。精漿成分を洗い落とした洗浄精子に対してSgIIが結合していないことを確認した後、FITC-SgIIを反応させた後,蛍光顕微鏡を用いて観察すると,ミトコンドリアを多量に含む精子の中部に特異的に結合することが確認された.結合する蛋白については現在解析中である.(2) 男性不妊症患者の精液中のSgII濃度およびPCI濃度と,病態との関連性を解析した.精嚢低形成患者の精漿中にはPCIはほとんど検出されなかった.また,精漿中にはPCI濃度はフルクトースに比較して射精後48時間を経過しても安定しており,精嚢機能不全あるいは射精管閉塞患者の診断に有用であると考えられた.(3) 精子無力症の患者の精漿中PCI濃度は高くPSA濃度は低いことから、PCIが精子自体に作用し精子運動能を抑制しているか、精子の運動能を高めるプロテアーゼの活性を阻害して、精子運動能を低下させている可能性があり現在解析中である。

Analysis of the interaction between SgII and PCI. Objective: To analyze the physiological function, determine the concentration of SgII and PCI in seminal plasma of healthy people and infertile men, and discuss the relationship between pathological factors. (1)Sperm washing and FITC staining were used to detect SgII(FITC-SgII) binding, and SgII binding was observed under microscope. After washing the seminal plasma components, FITC-SgII was detected by light microscopy, and the specific binding between the sperm and the central part of the sperm was confirmed. The binding protein was analyzed. (2)SgII concentration in semen of men with pregnancy disorders is higher than PCI concentration, and the relationship between disease and disease is analyzed. In the seminal plasma of patients with low semen formation, PCI is not allowed. The concentration of PCI in seminal plasma is useful for diagnosis of ejaculatory duct redness 48 hours after ejaculation. (3)In patients with asthenospermia, PCI concentration in seminal plasma is high, PSA concentration is low, PCI inhibits sperm motility, sperm motility is high, sperm motility is low, and the possibility of sperm motility is low.
