本研究は、骨吸収促進因子IL-6とIL-1βならびに骨吸収抑制囚子IL-4の産生における年齢的変化が辺縁性歯周炎による骨吸収の病態と進行にどのように影響を及ぼしているかを、RT-PCR法と免疫組織化学的染色法を用いて検討するものである。平成9年度は、予備実験を行った。平成10年度は、歯槽骨の吸収を伴う進行性の歯周炎が認められる成人患者と仮性ポケットを伴うような高度な歯周炎が認められる若年者から外科処置時に得られた歯肉組織を用いた。歯肉片よりAPGC法の変法により全RNAの抽出し、RT-PCRを行った。IL-1β、IL-4、IL-6の特異的プライマーとpositive controlとしてハウスキーピングジーンであるG3PDHを用いた。増幅されたDNAの検出は4%アガロースゲルで電気泳動し、UVにて観察した。今回の検索ではIL-1βが若年者においてその発現が高い傾向が認められたが、有為ではなかった。IL-4、IL-6に関しては有為な差は認められなかった。免疫組織学的染色については凍結歯肉片から連続切片を作成し、酵素抗体法(ABC法)を行った。IL-1β、IL-4、IL-6に関して有為な差が認められなかったため、他の抗体を使用して検討した。その結果、細胞周期関連タンパク質であるp21に関して若年者の歯肉増殖症でその発現が多い傾向が認められた。

In this study, we investigated the effects of bone resorption promoter IL-6, IL-1β and bone resorption inhibitor IL-4 on bone resorption pathologies in periodontitis, and RT-PCR immunohistochemical staining. 9 years ago, I was prepared to go. IL-1β has a high tendency to be detected in the present study. IL-4 and IL-6 are not suitable for use. Immunohistochemical staining was performed using the enzyme antibody method (ABC method). There are many differences in the relationship between IL-1β, IL-4, and IL-6, and the use of other antibodies has been studied. As a result, the cell cycle is related to p21, which is related to the occurrence of reproductive diseases in young people.
