项目摘要

現在治療抵抗性とそうでない慢性C型肝炎のCORE蛋白とNS5A蛋白をHuh7,HepG2,Hep3Bといった肝細胞癌由来のセルラインにトランスフェクトしており、細胞に遺伝子を移入するベクターが恒常的に細胞に発現していることを確認している。現在CORE蛋白とNS5A蛋白がどの程度実際に発現しているかウェスタンブロッティング法にて定量中である。今後インターフェロントランスダクションに関連した蛋白の発現を、mRNA段階ではCATアッセイもしくはルシフェラーゼアッセイで確認する。この際インターフェロンの影響を確認するため段階濃度でそれぞれの抗ウイルス蛋白の発現を測定する。また同時にNS5A蛋白がどの様な機序で細胞内で作用しているかを明らかにする。またこれらのHCV蛋白がインターフェロン投与の際ウイルス排除に重要な役割を果たしていると考えられている2′,5′-OAS, PKR, Mx蛋白等の抗ウイルス物質が産生されるが、現在NS5A蛋白がPKRを直接阻害することがin vitroの実験で示されておりこれがインターフェロン治療抵抗性の重要な機序の一つであると考えられている。既に我々は2′,5′-OAS,がウイルスのNS5蛋白の影響を強く受けていることをヨーロッパの雑誌やアメリカでの学会で発表してきたが現在肝癌細胞株にNS5A蛋白を発現させ、どの様な機序で抗ウイルス蛋白である2′,5′-OASやPKRが抑制されるか検討中である。

The HCV protein is a direct inhibitor of PKR, such as 2′, 5 ′-OAS, PKR, Mx protein, etc., which is an important mechanism of therapeutic resistance. In addition, the expression of NS5A protein in hepatoma cell line NS5A was strongly influenced by 2′,5 ′-OAS and PKR inhibition.
