ラット消化管のプロエンケファリンA(PENK A)、プロエンケファリンB(PENK B)、およびプロオピオメラノコルチン(POMC)などのmRNA量の発達に伴う変化を研究した。生後5、10、15、20、30、40、および70日目のラットの食道、胃、小腸、盲腸および結腸などを摘出し、グアニジンチオシアネート・塩化セシウム法で全RNAを抽出し、オリゴdTセルロースカラムを用いmRNAを得た。このmRNAを用いアガロースゲル電気泳動を行った後、mRNAをナイロン膜へブロッテングした。そして、3種のオピオイドペプチド前駆体タンパク質mRNAの特定の部位に対応した合成オリゴヌクレオチドの3′末端を^<32>P-alphaATPでテーリングしてプローブとして用い、ハイブリダイゼーション後、-70℃でオートラジオグラフィーを行い、mRNAのバンドを検出するとともに、バイオイメージアナライザーを用いmRNAに相当するバンドの定量を行った。その結果、胃の単位全mRNA量当たりのPENK A mRNA量は、生後70日目に比べ、生後5、10および15日目では4倍前後多く、その後20日目では2倍前後、30および40日目では1.5倍前後となった。PENK B mRNA量の発達に伴う変化も類似していた。一方、POMC mRNA量は、生後70日目に比べ、生後5、10および15日目の方が少なく、20、30および40日目でほぼ同じになった。すなわち、前回までに明らかにした全脳のmRNA量の変化は、POMCでは類似していて(両者とも発達に伴い徐々に増加する)、PENK AおよびBでは著しく異なる(全脳では徐々に増加するのに対して、胃では5〜15日目に多く、20日目に急激に減少し、その後徐々に減少する)ことが明らかにされた。この原因を目下検討中である。また、胃以外の部位の発達に伴う変化も研究中である。
