Antiganglioside antibodies are found in the axonal form of Guillain-Barre syndrome and multifocal motor neuropathy. Several studies indicated that they may produce conduction block through Na channel inactivation. To confirm this contention, we devised a method to analyze saltatory conduction in single myelinated fiber under the influence of the sera. Despite the premise, we found conduction block in only 2 out of 38 fibers examined with high-titer anti-GM1 rabbit serum. None showed conduction block by human sera. Only one out of 25 exhibited block after exposure to lymphokine produced by lymphocytes incubated with the anti sera. These finding do not support the view that these sera play an important role in the pathogenesis of these diseases.
